Dear Experts, please let me know how we can automate all files from D drive to SharePoint or one drive, without creating a backup copy in my PC.
You can use the File System "Get File Content" actions and then "Create" in SharePoint.
You will need to install a gateway to use the File System set of actions.

HOWEVER.
I don't recommend using Power Automate for this purpose. Use a migration tool instead.
