First Question
Does the user is able to access and modify Powerapps's global variable? Because I have a variable that stores the user's role for restricting action and access.
Second Question
The users are possible to navigate to other screens if I do not provide any navigation buttons to them? Or could you guys give me a suggestion to restrict particular role accesses to some screens?
If you don't code a way for them to edit a variable, they can not edit a variable, same for screen navigation.

A variable is unique to the user's session and they can only modify it if you give them the ability with triggered code to do this (on a button for instance) - so in general the answer is no. Second item same answer - you can control what a user can do.
