Hi All,
I am sending an email to the manager saying that request is created with all the data from sharepoint
Again the manager is coming to Share point or power apps and assigning the member (kind of updating a filed)
Is the any option in email the Manager can Assign the user for the created Request ?

Well, it depends.
You could - in the Start & Wait for an Approval - use the Response Options to instead put in assignee names. It depends how many you have, and how often they change.
You could - in Send An Email with Options - put the possible assignees into the User Options. But that's not the same as an approval, it won't have reminders / stay in a specific spot (aside from email) unless you build it in.
You could use an adaptive card in Microsoft Teams to create a dropdown list of people to choose from.
The first will be the easiest, by far. It depends what you're willing to get into.
